These plexinD1flox mutant mice possess loxP sites flanking exon 1 of the plexin D1 (Plxnd1) gene. PlexinD1 is a transmembrane receptor for class 3 semaphorins expressed prominently in developing vascular endothelial cells. Expression is also seen in the central nervous system, in the salivary gland, neural crest and in bone during embryogenesis. PlexinD1 has been found to play a role in the migration and proliferation of some vascular tumor cells. Homozygous plexinD1flox mice are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 1 deleted in the cre-expressing tissues, abolishing gene function.
For example when bred to B6.Cg-Tg(Tek-cre)1Ywa/J mice
(Stock No. 008863), expressing cre-recombinase in the vascular endothelium, these mice exhibit congenital heart, vascular and skeletal defects.
When bred to B6.Cg-Tg(CAG-cre/Esr1*)5Amc/J mice
(Stock No. 004682), expressing tamoxifen-inducible cre-recombinase, these mice exhibit alterations in retinal vasculature.
A targeting vector was designed to insert a frt-flanked neomycin resistance (neo) cassette and a loxP site upstream of exon 1, followed by a second loxP site downstream of exon 1 of the plexin D1 (Plxnd1) gene. The construct was electroporated into 129Sv/Ev-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric mice were bred to C57BL/6J mice. The resulting offspring were bred with B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ transgenic mice (see Stock No. 009086) to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. These mice were backcrossed to C57BL/6J mice for at least 3 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
