Detailed description

XRCC2 is a member of the RecA/RAD51-related protein family associated with repair of double stranded breaks using homologous recombination. In this strain, the ?Cre-lox? system is used to remove exon 3, which contains 75% of the open reading frame (ORF). Most homozygotes die during early embryogenesis (E9-E10) due to apoptosis of proliferating cells. embryonic fibroblasts (MEFs) derived from homozygous embryos are hypersensitive to DNA and exhibit spontaneous chromosomal rearrangements. This mutant mouse strain may be useful in studies of DNA repair in neural development.

Development

A targeting vector was designed to insert a loxP-flanked PGKneo cassette upstream of exon 3 and a loxP site downstream of exon 3. Exon 3 contains 75% of the open reading frame (ORF). The construct was electroporated into 129S1/Sv-dervied W9.5 embryonic stem (ES) cells. Transient Cre expression in targeted cells excised the neo cassette leaving a single loxP site. Correctly targeted ES cells were injected into blastocysts.The resulting chimeric animals were crossed to C57BL/6 mice and maintained by intercross.
