mThy1-hSNCA transgenic mice express the human α-synuclein gene under the direction of the mouse thymus cell antigen 1 promoter. Hemizygotes are viable, fertile, and normal in size. This line carries 1-2 copies of the transgene which is integrated on mouse Chromosome 11 (40456787-40495044) and results in a 38.4 kb deletion (PMID 32170061). These mice may be useful for studying Parkinson's disease pathogenesis and neurodegeneration elicited by the toxic effects of aggregation and somatic accumulation of SNCA.
The mThy1-hSNCA transgene consists of the human α-synuclein (SNCA) gene driven by a mouse thymus cell antigen 1 (Thy-1) promoter. The construct was microinjected into fertilized C57BL/6NTac oocytes and males from founder line 15 were bred to C57BL/6NTac females to establish the colony. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6NJ inbred mice (Stock No. 005304) for at least one generation.
Analysis of the integration site indicates insertion occurred at Chr11:40456787-40495044 and resulted in a 38.4 Kb deletion (PMID 32170061).
