Description

This compound mutant strain combines the immunodeficiency features of the Prkdc^scid mutation and widespread fluorescent cell labeling capabilities of CAG-DsRed on the NOD genetic background. These mice are effective recipients of allogeneic and xenogenic grafts, and are an excellent source of fluorescent-labeled cells for transfer experiments.

Detailed description

These compound mutant mice carry both the Prkdc^scid (scid) targeted mutation and a transgene with a red fluorescent protein variant (DsRed*MST) under the control of a chicken beta-actin promoter and cytomegalovirus enhancer (CAG).

Mice homozygous for the DsRed transgene exhibit a robust and widespread expression of red fluorescent protein in all cells of embryos and adults. Animals homozygous for scid and hemizygous for DsRed are viable and fertile and exhibit the same immunodeficient characteristics of the parental NOD Prkdc^scid strain (e.g. T, B, and NK cell deficiencies - see Stock No. 001303).

These mice can act as recipients of allogeneic and xenogeneic grafts and perhaps more importantly, donors of fluorescent-labeled cells, making them an ideal model for cell transfer experiments.

Development

A CAG-DsRed*MST transgenic mouse (Stock No. 005441) was intercrossed with NOD background Prkdc^scid (Stock No. 001303) animals for more than 10 generations by the donating laboratory.

Molecular note: This strain is a derivative of Tg(ACTB-Bgeo,-DsRed*MST). A transgenic construct containing a Red Fluorescent Protein gene (DsRed.MST) under the control of the chicken beta actin promoter coupled with the cytomegalovirus (CMV) immediate early enhancer upstream of a loxP site flanked beta-geo gene and polyadenylation sites, was introduced into 129S6B6F1 derived G4 embryonic stem (ES) cells. Then the expression of lacZ was replaced with red fluorescent protein following in vitro Cre recombination. All tissues of homozygotes fluoresce red. Mice hemizygous for the transgene express red fluorescent protein less intensely than homozygotes. Expression is observed throughout all embryonic and adult stages and very high expression is found in pancreas, skeletal muscle, heart and seminal vesicle.
General note: Erratum publ Genesis 42:218 (2005) correction to original (2004) publication: The red fluorescent protein used is DsRed.MST, not DsRed.T3 as J:102837. DsRed.MST differs from DsRed.T3 in two amino acids. It has Alanine at positions 145 and 217.
