YFP and Cre are expressed from the basophil-specific Mcpt8 (mast cell protease 8; also known as Basoph8) promoter in this targeted mutation strain. This strain is useful for labeling, sorting and deleting basophil cell populations.

This targeted mutant strain expresses both yellow fluorescent protein (YFP) and Cre recombinase from the Mcpt8 (mast cell protease 8; also known as Basoph8) promoter. The YFP reporter labels MCPT8-expressing cells, enabling lineage marking of basophils as well as cell sorting by flow cytometry. Cre excision of loxP-flanked sequences is basophil-specific when these mice are bred with a floxed animal. Expression of the endogenous Mcpt8 gene is blocked, but basophil functions are not impaired.
When crossed with a strain expressing a floxed stop-diptheria toxin α from the Gt(ROSA26)Sor promoter (e.g. Stock No. 009670), selective deletion of basophils can be achieved.
Genomic sequence of the rabbit β-globin gene, enhanced yellow fluorescent protein (YFP), IRES, humanized Cre (mammalian-optimized), bovine growth hormone poly (A), and a loxP-flanked neomycin cassette were introduced to the mouse Mcpt8 (Basoph8) gene immediately after the 5' promoter and UTR. 129-derived embryonic stem cells encoding Prm-Cre were used to create the mutation and excise the neomycin resistance cassette. This strain was backcrossed to BALB/cJ for 10 generations by the donating lab.

Molecular Note | A 4.8 kb reporter cassette containing YFP, IRES site, humanized cre recombinase, bGH polyA, and a loxP-flanked neo selection marker was inserted at the Mcpt8 start site via homologous recombination in ES cells contain PrmCre (PC3 cells with Tg(Prm-cre)70Og). Upon crossing male chimeras to wild-type females, the loxP-flanked neo was deleted from the male germline by cre recombination, producing the final knock-in allele. |
