Description

The 91-1 mES cell line was used to generate Tc1 mice (Stock No. 010801 Tc(HSA21)1TybEmcf), which carry the 42Mb freely segregating human fragment of Chromosome 21 Hsa21 containing 269 genes, including most of the gene orthologs located on mouse Chromosome 10 (Mmu10), Mmu16, and Mmu17, which have been found to contribute to human Down Syndrome.

Detailed description

The 91-1 mES cell line was used to generate Stock No. 010801 B6;129S-Tc(HSA21)1TybEmcf/J, which provides a model of Down Syndrome in which only the female carriers consistently transmit the mutation through the germline. Mice of that stock carry the 42Mb freely segregating human transchromosomal segment containing 269 human genes, including most of the gene orthologues located on mouse Chromosome 10 (Mmu10), Mmu16, and Mmu17, which have been found to contribute to Down Syndrome.

Development

The 91-1 transchromosomal Hsa21 embryonic stem cell line was established through irradiation microcell-mediated chromosome transfer (XMMCT) of a 129S2 ES cell line. Using XMMCT, human Chromosome 21 (Hsa21) was allowed to freely segregate into the female 129S2/SvPas derived ES cell line, MPI VI. It was estimated that 91-1 contains at least 42 Mb (90%) of the 46.9mb Hsa21, or 92% of all known Hsa21 genes. This ES cell was used to generate the strain B6129S-Tc(Hsa21)1TybEmcf/J (Stock No. 010801).
