These Dmp1-Topaz transgenic mice express the topaz variant of GFP under the direction of the mouse Dmp1 promoter. Expression is observed in adult osteocytes of trabecular and metaphyseal regions of long bones, and in the cortical area of the femur and calvarial bone.

Dmp1-Topaz transgenic mice express the topaz variant of green fluorescent protein (GFP) under the direction of the mouse dentin matrix protein 1 (Dmp1) promoter. The donating investigator reports that homozygotes are viable. DMP1 is expressed in diverse cells within bone and tooth tissue and is required for bone and dentin mineralization. Under control of Dmp1, these mice express topaz protein in adult osteocytes of trabecular and metaphyseal regions of long bones, and in the cortical area of the femur and calvarial bone. Some topaz expression is also seen in brain and kidney.
The Dmp1-Topaz transgene was designed with the topaz variant of green fluorescent protein (GFP) driven by 7892 base pairs of the promoter, exon1, and part of exon 2 of the dentin matrix protein 1 (Dmp1) gene. The transgene was microinjected into fertilized C57BL/6 oocytes. Offspring were bred to C57BL/6J mice and founder line 1 was established. These mice were maintained on a C57BL/6J background. Upon arrival at The Jackson Laboratory, transgenic mice were bred to C57BL/6J inbred mice (Stock No. 000664) to establish the colony.

We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
