Mice hemizygous for the BAC Col10a1-mCherry transgene are viable and fertile with mCherry expression directed by the collagen, type X, alpha 1 (Col10a1) promoter/enhancer regions within the BAC transgene. COL10A1 is expressed in mature hypertrophic chondrocytes during endochondral ossification, but is absent in immature chondrocytes. These BAC mice express the mCherry protein in hypertrophic chondrocytes. Specifically, at E14.5 mCherry fluorescence is seen in the ribs adjacent to the spine and at the center of the cartilaginous anlagen in the developing forearm. At E17.5, Col10a1-mCherry expression is seen in zones of cartilage maturation in the appendicular, axial skeleton, and cranial base.
