Description

These Dgat1^flox mutant mice may be useful in generating conditional mutations for studying retinoid homeostasis, lipid metabolism and obesity.

Detailed description

These mice possess loxP sites on either side of exons 14 through 17 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 14 through 17 deleted in the cre-expressing tissue(s).

When bred to a strain with Cre recombinase expression in skin (see Stock No. 004782 for example), this mutant mouse strain may be useful in studies of retinoid homeostasis.

Development

A FRT site flanked targeting vector containing a GK-NEO cassette and a loxP site was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 17 of the targeted gene, and another loxP site was inserted upstream of exon 14. This construct was electroporated into unspecified 129S4/SvJae derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to C57BL/6 mice, and then crossed to an Actin-FLPe strain, on the C57BL/6J background, to excise the FRT flanked GK-NEO cassette. The mice were then backcrossed to C57BL/6J for 10 generations, and no longer carry the Actin-FLPe transgene. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
