These floxed mutant mice possess loxP sites flanking exon 3 of the Gtf2i gene. This strain may be useful for generating conditional mutations in applications related to embryonic and craniofacial development, and neural tube closure.

The targeted Gtf2i gene encodes the TFII-I phosphoprotein transcription factor and calcium channel regulator. Mutations in this gene have been associated with Williams-Beuren syndrome.
These mice possess loxP sites on either side of exon 3 of the targeted Gtf2i gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues.
Removal of the floxed sequence creates a null allele.
When bred to a strain with germline Cre recombinase expression (see Stock No. 004302 for example), this mutant mouse strain may be useful in studies of embryonic development, specifically neural tube and craniofacial development.
A targeting vector containing a FRT-site flanked PGKneo selection cassette was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 3 of the targeted gene, and another loxP site was inserted upstream of exon 3. This construct was electroporated into (129S6/SvEvTac x C57BL/6J)F1
derived D2 embryonic stem (ES) cells. Chimeric mice were generated by aggregating correctly targeted ES cells with CD-1 morula-stage embryos. The chimeric male mice were bred to ROSA26-Flpe female mice on a congenic C57BL/6J background (see allele in Stock No. 003946) to achieve germline transmission and remove the FRT site flanked PGKneo cassette. The mice were then crossed to C57BL/6J mice for 3 generations to breed out the ROSA26-Flpe allele.
Heterozygotes were crossed to generate homozygotes.
Homozygous embryos were cryopreserved. To establish our live colony, the line was recovered.
