In this strain, the insertion of a ROSAFARY gene trap vector creates a null allele in the Fbxl3 (F-box and leucine-rich repeat protein 3) gene. Homozygotes have a free running period of 27 hours as measured by running wheel behavior (23.7 hours in wildtype controls).
The ROSAFARY gene trap vector, incorporating a beta galactosidase/neomycin (βgeo) cassette and an FRT-flanked PGK-hygromycin cassette, was introduced to 129S4/SvJaeSor-derived AK7.1 embryonic stem (ES) cells. G418-resistant clones were selected and sequenced, enabling the identification of gene trap cell line S13-12G1 carrying a disruption of the Fbxl3 gene. The gene trap cell line was introduced to C57BL/6J blastocysts. The resultant strain was backcrossed three times to C57BL/6J by the donating laboratory.
