These Il4/Il13flox mutant mice possess a loxP site upstream of exon 3 of the interleukin 13 (Il13) gene and a second loxP site downstream of exon 4 of the interleukin 4 (Il4) gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have nucleotides between exons 3 of Il13 and exon 4 of Il4 deleted in the cre-expressing tissue. This strain may be useful for studying immune function in mice lacking Il4 and Il13.
A targeting vector was designed to insert a loxP-flanked blasticidin (bsd) resistance cassette upstream of exon 3 of the interleukin 13 (Il13) gene, and a frt-flanked neomycin resistance cassette followed by a single loxP site downstream of exon 4 of the interleukin 4 (Il4) gene. The construct was electroporated into 129P2/OlaHsd-derived E14 embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a pMC-CreN expression plasmid to delete the bsd cassette. Resulting ES cells contained multiple gene rearrangments; intact floxed-exons, intact floxed-bsd cassette, or excision of both the exons and the bsd cassette. Correctly targeted ES cells, containing only the floxed-exons, were injected into C57BL/6 blastocysts and resulting chimeric males were bred with C57BL/6 females. Mice heterozygous for the Il4/Il13flox allele were bred to mice containing Gt(ROSA)26Sortm1(FLP1)Dym to delete the neo cassette. This line was backcrossed to BALB/c mice for at least 10 generations. Upon arrival at The Jackson Laboratory Repository, mice were bred to BALB/cJ inbred mice (Stock No. 000651) for at least one generation.
