In this strain, a neomycin resistance (neo) cassette replaces exon 1 and part of exon 2 of the p21 protein (Cdc42/Rac)-activated kinase 6 (Pak6) gene, abolishing gene expression. Homozygotes are viable, fertile, normal in size, and do not display any gross physical abnormalities. Pak6 is a member of the group B family of PAK serine/threonine kinases and is expressed in the cortex, striatum, and hippocampus. It is involved in neuronal growth and cell survival. The absence of an apparent aberrant phenotype in these mice may be due to the compensatory actions of Pak7 (p21 protein (Cdc42/Rac)-activated kinase 7). When maintained on a background also lacking Pak7 activity, these mice have impaired cognitive function and deficits in learning and memory retention. These mice may be useful for studying learning, memory, and locomotion.
A targeting vector was designed to replace exon 1 and part of exon 2 encoding the p21 protein (Cdc42/Rac)-activated kinase 6 (Pak6) gene with a neomycin resistance (neo) cassette. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and the resulting chimeric males were bred to C57BL/6 females. The donating investigator reported that these mice were backcrossed at least nine generations to C57BL/6 background (see SNP note below). Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.
A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Four markers throughout the genome were segregating for 129, suggesting an incomplete backcross.
