These Pdx-Cre transgenic mice express Cre recombinase under the control of the mouse Pdx1 promoter and may be useful for generating pancreas-specific conditional mutations.

Mice hemizygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express Cre recombinase under the control of the mouse Pdx1 (pancreatic and duodenal homeobox 1) promoter. Mosaic Cre recombinase activity is detected in the pancreatic epithelium, antral stomach and duodenum in neonates and in pancreatic beta islet cells in adults. No Cre recombinase activity is detected ectopically to the Pdx1 expression domain. When crossed with a strain containing loxP site-flanked sequences, Cre-mediated recombination results in deletion of the floxed sequences in the cre-expressing tissues of the offspring. It is not known if homozygotes are viable.
A transgenic construct containing sequence encoding Cre recombinase under the control of a 4.5 kb promoter fragment of mouse Pdx1 and a human ACTB 3' UTR followed by a poly A signal was introduced into FVB/N donor eggs. In founder line 6 the trangene integrated on chromosome 8, 48 kb upstream of Zfp423. The integration site contains a 1.5 kb genomic duplication. The donating investigator stated that these mice were backcrossed for 15 generations onto the C57BL/6 background (see SNP note below). Upon arrival at The Jackson Laboratory Repository, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 1 of 27 markers throughout the genome still suggested FVB in the background, all 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a C57BL/6N genetic background.

When maintaining a live colony, these mice can be bred as hemizygotes. It is not known if homozygotes are viable.
