These EIIa-Cre transgenic mice express Cre recombinase under the control of the adenovirus EIIa promoter. When crossed with a strain containing loxP site-flanked sequences, Cre-mediated recombination occurs in a wide range of tissues, including germ cells that transmit the genetic alteration to progeny.

This line carries a Cre transgene under the control of the adenovirus EIIa promoter that targets expression of Cre recombinase to the early mouse embryo. Cre expression is thought to occur prior to implantation in the uterine wall. A mosaic pattern of expression is commonly observed. Cre-mediated recombination occurs in a wide range of tissues, including the germ cells that transmit the genetic alteration to progeny.

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
The EIIa-Cre plasmid was constructed by replacing the human cytomegalovirus promoter in pBS185 with the adenovirus EIIa promoter from pEII-lacZ. cre activity present in zygotes of the EIIa-cre genotype was measured as follows. EIIa-cre/EIIa-cre homozygous animals were mated with +/alphaA-loxP-STOP-loxP-TAg heterozygous animals and the presence of lens tumors in double transgenic progeny, indicative of cre-mediated excision of STOP sequences and a concomitant activation of SV40 TAg, was scored. The allele originated on an FVB/N (Stock No. 003314) background prior to transfer to the NOD/ShiLtJ utilizing marker assisted technology. At generation N4, all markers outside of the Chr. 6 congenic region (SNPS 06-027868538-G, 06-042391222-G and 06-053482462-M) are of NOD origin.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
