Description

Detailed description

Tie2-B4galnt2 transgenic mice have beta-1,4-N-acetyl-galactosaminyl transferase 2 (B4galnt2) expression being driven by the vascular endothelial-specific receptor tyrosine kinase (Tie2) promoter in vascular endothelial cells. Hemizygous mice are viable, fertile, and normal in size. These mice exhibit lower plasma von Willebrand factor (VWF) levels and prolonged bleeding time characteristic of RIIIS/J mice (Stock No. 000683), which contain a modified VWF (Mvwf) gene. These mice may be useful for studying plasma VWF levels, cohesion and aggregation of platelets at the site of injury, bleeding, and clotting associated with human von Willebrand disease.

Development

A transgenic vector was generated to include the endothelial-specific receptor tyrosine kinase (Tie2) driving vascular endothelial specific expression of beta-1,4-N-acetyl-galactosaminyl transferase 2 (B4galnt2), followed by a simian virus 40 intron and a polyadenylation signal (polyA). This transgene is lacking 23 bp of B4galnt2 3'UTR for detection purposes. The construct was microinjected into (C57BL/6J x SJL)F1 fertilized oocytes, and mice from founder line 1108 were bred to C57BL/6J. These Tie2-B4galnt2 mice were backcrossed to C57BL/6J for at least 23 generations to establish a hemizygous colony. Upon arrival at The Jackson Laboratory, transgenic mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
