These Il6ra^fl/fl mutant mice possess loxP sites flanking exons 4-6 of the interleukin 6 receptor alpha chain (Il6ra) gene. This strain may be useful for studying the role of Il6 in immune response, epithelialization, angiogenesis, macrophage infiltration, and wound healing.

Detailed description

These Il6ra^fl/fl mutant mice possess loxP sites flanking exons 4-6 of the interleukin 6 receptor alpha chain (Il6ra) gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When bred to mice that express tissue-specific Cre recombinase, resulting offspring lack detectable Il6ra in the cre-expressing tissues. This strain may be useful for studying the role of Il6 in immune response, re-epithelialization, angiogenesis, macrophage infiltration, and wound healing.

For example, when bred to a strain expressing Cre recombinase in the myeloid cell lineage (see Stock No. 004781 for example), this mutant mouse strain may be useful in studies of inflammation and wound healing.

When bred to a strain expressing Cre recombinase in liver (see Stock No. 003574 for example), this mutant mouse strain may be useful in studies of acute phase response.

Development

A targeting vector was designed to insert a loxP site upstream of exon 4, and a frt-flanked neomycin resistance (neo) cassette followed by a second loxP site downstream of exon 6 of the interleukin 6 receptor alpha chain (Il6ra) gene. The construct was electroporated into C57BL/6N-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J-Tyr^c-2J/J (Stock No. 000058) blastocysts and resulting chimeric mice were bred with B6;SJL-Tg(ACTFLPe)9205Dym/J mice (Stock No. 003800) to delete the neo cassette. These progeny were then crossed to C57BL/6 for more than 3 generations to remove the FLP recombinase, resulting in a colony homozygous for the floxed-Il6ra (Il6ra^fl/fl) allele. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
