These Acf7 floxed mutant mice may be useful in generating conditional mutations for studies of the cytoskeleton and early embryonic development.

These mice possess loxP sites on either side of exons 6 and 7 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 6 and 7 deleted in the cre-expressing tissue(s).
A targeting vector containing Macf1 sequence including exons 6 and 7, neomycin resistance and herpes simplex virus thymidine kinase genes was utilized in the construction of this mutant. A loxP site was placed downstream of exon 7 and another loxP site and the Neo selection cassette was inserted upstream of exon 6. The construct was electroporated into 129Sv-derived embryonic stem (ES) cells. The resulting chimeric male animals were backcrossed to wildtype C57BL/6 mice. Upon arrival at The Jackson Laboratory the mice were crossed to C57BL/6J mice at least once to establish the colony.
