These Pax7f floxed mutant mice may be useful in generating conditional mutations for studying muscle regeneration.

These mice possess loxP sites on either side of exon 2 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissue(s).
For example, when crossed to a strain expressing tamoxifen-inducible Cre recombinase in myogenic progenitor cells (see Stock No. 012476), this mutant mouse strain may be useful in studies muscle stem cells.
A PGK-Neo selection cassette flanked by FRT sites was inserted downstream of exon 2 of the targeted gene. A loxP site was inserted downstream of the selection cassette. Another loxP site was inserted upstream of exon 2. The targeting construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+ derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to mice expressing FLP recombinase under the control of an Actin promoter (on the C57BL/6 background) to remove the neo selection cassette. The mice were then intercrossed for 4 generations. Upon arrival at The Jackson Laboratory the mice were crossed to C57BL/6J once to establish the colony.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
