Description

The serine 831 phosphorylation site was mutated to alanine in this Gria1 (glutamate receptor, ionotropic, AMPA1 (alpha 1)) targeted mutation strain. Mutant protein is expressed in the brain. Appetitive incentive learning is defective in these mice, but performance of a number of other measures of motivated performance are unaffected. These mice may be useful in studies of normal and maladaptive forms of motivated behavior (e.g. addiction and eating disorders).

Detailed description

The serine 831 phosphorylation site was mutated to alanine in this Gria1 (glutamate receptor, ionotropic, AMPA1 (alpha 1)) targeted mutation strain. Normal GRIA1 protein expression is replaced by the mutant form in the brain. Normal basal synaptic transmission and plasticity are seen in young (3 week old) and adult (more than 3 month old) mice. Appetitive incentive learning is defective in these mice, but performance of a number of other measures of motivated performance are unaffected. These mice may be useful in studies of normal and maladaptive forms of motivated behavior (e.g. addiction and eating disorders).

Development

A Ser831Ala (TCC -> GCC) mutation was introduced to exon 17 of the targeted gene using (129X1/SvJ x 129S1/Sv)F1- Kitl⁺-derived R1 embryonic stem (ES) cells. Heterozygous mice were bred to CMV (cytomegaolovirus)-promoter cre mice to excise a floxed neomycin resistance cassette used as a selection marker. Cre was bred out in the next generation and this strain was backcrossed thirteen times to C57BL/6 by the donating laboratory.
