Description

This BAC transgenic line expresses CreERT under the control of the Slc1a3 (solute carrier family 1 (glial high affinity glutamate transporter); GLAST) promoter. When crossed with a strain containing a loxP site flanked sequence, the offspring are useful for generating tamoxifen-induced, Cre-mediated targeted glia and neural progenitor cell-specific deletions. This strain is a useful tool in studies of neurodevelopment.

Detailed description

This BAC transgenic line expresses CreERT under the control of the Slc1a3 (solute carrier family 1 (glial high affinity glutamate transporter); GLAST) promoter. When crossed with a strain containing a loxP site flanked sequence, the offspring are useful for generating tamoxifen-induced, Cre-mediated targeted glia and neural progenitor cell-specific deletions. Injection of 4-hydroxytamoxifen leads to recombination specifically in Muller glia in the retina, in astrocytes of the brain, and in neural progenitors, including those that give rise to hippocampal neurons and olfactory neurons in the rostral migratory stream. This strain is a useful tool in studies of neurodevelopment.

Development

A CreER(T) (CreEsr(T)) coding region with a polyA site was inserted at the initiator methionine codon of Slc1a3 (GLAST) contained in a >100 kb BAC. The vector was introduced to B6SJLF1 embryos and the line was backcrossed approximately 5 times to C57BL/6 by the donating laboratory. A residual FRT site preceding the coding region remains from other genetic manipulations.

A 32-SNP (single nucleotide polymorphism) panel analysis performed by The Jackson Laboratory revealed that this strain is on a mixed genetic background (17 out of 32 markers are segregating for non-C57BL/6 alleles).
