This Chromosome 4 congenic stock contains the C57BL/10SnJ derived Idd9.1 region introgressed into the NOD/MrkTac background and exhibits significantly reduced diabetes incidence. This strain may be useful for positional mapping of the Idd9.1 loci and studying candidate genes within the Idd9.1 region.

Fine mapping performed at The Jackson Laboratory indicates the congenic interval to be as published. However, a spontaneous mutation was identified in this colony. Mice exhibited early non-Type 1 diabetes onset with 100% of the females diabetic by 18 weeks of age and 36% of the males diabetic by 8 weeks of age. After 2 backcrosses with Stock 1976 - NOD/ShiLtJ prior to making homozygous it appears that we have bred away from the mutation.
This NOD/MrkTac congenic stock, commonly referred to as NOD.B10-Idd9R1565 or NOD.B10-Idd9.1), carries C57BL/10SnJ (Stock No. 000666) derived genome on Chromosome 4 containing Idd9.1 region. This congenic interval wholly contains the 8Mb region defining Idd11 described by Brodnicki et al, 2000. The Idd9.1 region contains 329 genes, including candidate gene Lck.
This strain exhibits significantly diminished diabetes development when compared to NOD controls; with approximately 36% of the females becoming diabetic by 200 days of age. NOD-Idd9.1 mice are significantly more protected from diabetes development than NOD.B10-Idd9.3 (Stock No. 012311). NOD.B10-Idd9.1 exhibit enhanced development and function of CD4+CD25+ regulatory T cells than NOD controls. Data suggests that the Idd9.1 region regulates the suppressive activity and anti-diabetogenic potential of FoxP3+CD4+CD25+ Treg cells.
This strain may be useful for positional mapping of the Idd9.1 loci and studying candidate genes within the Idd9.1 region.
This NOD congenic stock contains a genomic segment derived from C57BL/10SnJ (Stock No. 000666) introgressed into the Idd9.1 region of Chromosome 4. This congenic region contains 329 annotated genes, including candidate gene Lck. This strain was produced by mating mice from NOD/MrkTac to C57BL/10SnJ followed by 15 additional crosses (N16) to NOD/MrkTac with selection for Idd9.1 markers. Siblings carrying the C57BL/10SnJ derived 18.5Mb chromosomal segment denoted by markers D4Mit258 (119Mb) through D4Mit339 (133.9Mb) were intercrossed to make homozygous. 5kSNP Chip Data identifies no C57BL/10SnJ genetic contamination outside of the Chromosome 4 congenic interval. The Type 1 Diabetes Resource received this stock in 2010.
