This Myc-tagged Piwil4 (piwi-like homolog 4 (Drosophila); also known as Miwi2) transgenic line permits the biochemical purification of the Miwi2 complex (a key component of the piRNA pathway) using robust anti-Myc antibodies. Expression, under the control of the transgene's native promoter, reproduces the developmentally timed pattern and subcellular localization of the endogenous protein. It begins as early as embryonic day 14.5 (E14.5) in developing germ cells of the testes and persists until shortly after birth. This strain may be helpful in studies of stem cell differentiation and spermatogenesis.
Three Myc tags were inserted at the N-terminus of the mouse Piwil4 (Miwi2) gene in BAC RP23-414K6. The linearized BAC was injected into B6SJLF1 hybrid oocytes to generate transgenic animals. The donating laboratory maintained the line on a mixed background composed of C57BL/6, SJL and outbred CD1.
