Description

Full-length mutant human SLC6A4 cDNA replaces exons 2-6 of the mouse solute carrier family 6 (neurotransmitter transporter, serotonin), member 4 gene. The native mouse promoter drives expression. These mice do not demonstrate an observable phenotype when untreated, but may be useful in inhibitory drug studies of serotonin transporter (SERT) allosteric modulation and re-uptake.

Detailed description

Full-length mutant human SLC6A4 cDNA replaces exons 2-6 of the mouse solute carrier family 6 (neurotransmitter transporter, serotonin), member 4 gene, in this strain. The native mouse promoter drives expression. These mice do not demonstrate an observable phenotype when untreated, but may be useful in inhibitory drug studies of serotonin transporter (SERT) allosteric modulation and re-uptake.

Development

Full-length mutant (II/552,553/VT - MS/558,559/SN - SI/574,575/TT) human SLC6A4 cDNA replaces exons 2-6 of the targeted mouse gene. C57BL/6-derived embryonic stem (ES) cells were used to create the mutation and this strain was maintained on a C57BL/6 background by the donating laboratory (see SNP note below).

A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, 5 markers in 2 of 4 mice that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6J ; C57BL/6N genetic background.
