Description

This FLP deleter strain is a suitable alternative to, and complement with the Cre-loxP system for in vivo genetic engineering and may be useful for generating FRT-FLP conditional mutations. Under the Pgk1 promoter, expression is ubiquitous in all tissues examined with strongest expression in the testis and ovary.

Detailed description

These transgenic mice express the mouse codon-optimized FLP recombinase under the direction of the mouse Pgk1, phosphoglycerate kinase 1, promoter. Transgene expression is detected in testis, ovary, brain, heart, liver and muscle by RT-PCR. When crossed with a strain containing a FRT site-flanked sequence of interest, FLPo recombinase activity is detected in all cells with complete recombinase mediated excision of the target. Mice hemizygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. The Donating Investigator has not attempted to make the strain homozygous.

Development

A transgenic construct containing the mouse codon-optimized FLP recombinase, FLPo, under the control of the mouse Pgk1, phosphoglycerate kinase 1, promoter was injected into fertilized hybrid (C57BL/6 x C3H) mouse oocytes. Founder line 10 was subsequently established. The donating investigator reported that these mice were then backcrossed to C57BL/6J (see SNP note below) for 5 generations. Upon arrival at The Jackson Laboratory in 2010, sperm was cryopreserved. To establish our live colony in 2010, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Four of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating, including the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.

Molecular note: A transgenic construct was created with the Pgk1 promoter driving mouse codon-optimized FLP (FLPo) recombinase. The linearized construct was injected into fertilized C57BL/6 x C3H oocytes. Six founder mice showing germline transmission were generated. These animals demostrated FLP expression as early as 3.5 embryonic days and adults displayed ubiquitious expression in all tissues examined with strongest expression in the testis and ovary. Line 10 showed 100% recombination efficiency when crossed to mice expressing a Ghr allele with a FRT-flanked PGK-Neo cassette in intron 5. (This Ghr allele is the parent construct of the Ghr^tm1.1Masp allele.)
