This transgenic line expresses hepsin and hrGFP in the ventral lobe of the prostate and exhibits disruptions in the basement membrane in mature males. This mutant mouse strain may be useful in studies of prostate carcinogenesis and metastasis.

Hemizygous mice are viable and express mouse hepsin (Hpn) and humanized Renilla Green Fluorescent Protein under the control of the rat probasin promoter. Tissues in the transgenic animals show GFP fluorescence patterns consistent with the hepsin transgene expression, specifically in the ventral lobe of the prostate. Immunofluorescence staining with antibodies specific to basement membrane, histological and electron microscopical examination suggest that the basement membrane in the prostate gland is severely compromised. This mutant mouse strain may be useful in studies of prostate carcinogenesis and metastasis.
When crossed with mice carrying the LBP-Tag line 12T-7f, Tg(Pbsn-Tag)12T7fRjm transgene (Kasper S et al., Lab Invest. 1998 Jun;78(6):i-xv), 20% of the resulting double transgenic mice (at 21 weeks of age) develop prostate cancer metastasis in liver, lung and bone.
A transgenic construct containing a modified rat probasin (Pbsn) promoter, beta globin intron, mouse hepsin (Hpn) full length cDNA , and an IRES-humanized Renilla (hr)GFP cassette was injected into C57BL/6 X CBA fertilized embryos. Founder line D was subsequently established. This line has been backcrossed to C57BL/6 for at least 6 generations.
In May 2010, a single nucleotide polymorphism (SNP) assay predicted these mice were 75% congenic onto the C57BL/6 genetic background (8 of 32 markers were not C57BL/6-allele type (on chr. 3, 5, 6, 8, 11, 12, 13, and 19)).
