Description

This transgenic line expresses a portion of human chromosome 22q11.2, a region related to cognitive impairment. The construct includes TXNRD2, COMT, and ARVCF. Mice display impaired working memory and enhanced incentive learning and may be useful in the study of learning and memory.

Detailed description

Transgenic mice are viable, fertile and normal in size. This transgenic line expresses a portion of human chromosome 22q11.2, a region associated with cognitive impairment. The construct includes thioredoxin reductase 2 (TXNRD2), catechol-o-methyltransferase (COMT), and armadillo repeat gene deleted in velo-cardio-facial syndrome ( ARVCF). Mice exhibit a shorter latency to reach a rewarded goal (enhanced incentive learning), but by two months, performed more poorly than wild-type mice in delayed rewarded alternation, suggesting an impairment in prolonged maintenance of working memory. This mutant mouse strain may be useful in the study of learning and memory.

Development

The transgenic construct consists of bacterial artificial chromosome (BAC) clone 467.8 encoding an approximately 190 kb segment from human chromosome 22q11.2 including TXNRD2, COMT, and ARVCF. Transgenic mice were generated by microinjection into FVB mouse embryos. This line has been backcrossed to C57BL/6 for at least 10 generations by the donating investigator.
