These Gfap-EGFP transgenic mice have EGFP expression directed primarily to Muller cells and may be useful in studies of retinal glial cells.

Mice harboring the Gfap-EGFP transgene are viable and fertile. The mouse glial fibrillary acidic protein (Gfap) promoter region directs expression of enhanced green fluorescent protein (EGFP) primarily to adult mouse retinal Muller cells. Developmental expression of EGFP mostly parallels GFAP in retinal astrocytes. Expression of EGFP first appears in Muller cells around two weeks of age. However, not all Muller cells express EGFP.
In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
The Gfap-EGFP transgene was designed with a 2.5 kb 5' fragment of the mouse Gfap glial fibrillary acidic protein placed upstream of an enhanced green fluorescent protein (EGFP) coding sequence and SV40 polyA sequence. This transgene was microinjected into fertilized ICR(CD-1) mouse oocytes. Founder mice (line 3739) were mated with ICR(CD-1) mice for several generations and subsequently bred to C57BL/6 mice for 15 generations prior to arrival at The Jackson Laboratory. The Donating Investigator indicates that homozygotes are viable, but maintained the strain by mating hemizygotes to outbred/inbred mice. Upon arrival, transgenic mice were bred with C57BL/6J to establish the colony.
SNP (single nucleotide polymorphism) analysis performed by The Jackson Laboratory revealed that the Donating Investigator used C57BL/6N (5 of 5 markers segregating) in the development of the strain.
