The NFAT-luc transgene has luciferase expression directed by a Myh6/NFAT hybrid promoter (composed of a minimal mouse alpha myosin heavy chain cardiac promoter [Myh6] with upstream copies of the nuclear factor of activated T cell binding site sequence [NFAT; from the mouse interleukin 4 (Il4) promoter]). In the hearts of adult NFAT-luc transgenic mice, NFAT-luc activity is upregulated during pathological cardiac hypertrophy induced by pressure-overload; more dramatically following myocardial infarction-induced heart failure. These NFAT-luc transgenic mice are a reporter of calcineurin-NFAT signaling, and may be useful in studying cardiac hypertrophy and calcineurin/NFAT coupling.

The mouse alpha myosin heavy chain cardiac promoter drives expression of nuclear factor of activated T cell (NFAT) binding sites and a luciferase reporter to detect calcineurin-NFAT signaling. In the adult heart, NFAT-luciferase activity is upregulated in association with cardiac hypertrophy induced by pressure-overload and following myocardial infarction-induced heart failure.
Nine copies of a nuclear factor of activated T cell (NFAT) binding site (5'-TGGAAAATT-3') from the interleukin 4 (Il4) promoter were placed 5' to a mouse alpha myosin heavy chain (Myh6) and upstream of a luciferase promoter. The transgene was created and maintained on an FVB genetic background by the donating laboratory.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
