Tnk1 (tyrosine kinase, non-receptor, 1) is considered to be a tumor suppressor that functions to down-regulate Ras activity. Both heterozygous and homozygous targeted mutant mice develop spontaneous tumors, including lymphomas and carcinomas, at high rates (27% and 43% respectively). Average latency is 17 months. On the C57BL/6 genetic background, the majority of homozygous mouse tumors, predominantly of B cell origin, occur in the small intestine and mesenteric lymph nodes of the gastrointestinal tract. Lymphomas also develop in the spleen, thymus, salivary gland, and pancreas.

In heterozygous mice, expression of the targeted gene is switched off in tumors due to hypermethylation of the promoter; surrounding tissues still express the gene. A large proportion of tumor-bearing heterozygous mice form lung and intestinal adenomas. Roughly 30% develop pancreatic islet cell hyperplasia. Malignant lymphomas occur at a lower frequency (28%) in spleen, pancreas, small intestine, and mesenteric lymph nodes in heterozygotes.
