These Dazl-eGFP transgenic mice have EGFP expression directed primarily to adult mouse testis in a developmentally-regulated, stage-specific expression pattern during spermatogenesis by the mouse Dazl (deleted in azoospermia-like) promoter/enhancer regions, and may be useful for gametogenesis studies (including pachytene spermatocytes).

Mice harboring the Dazl-eGFP transgene are viable and fertile, with the mouse Dazl (deleted in azoospermia-like) promoter/enhancer regions directing expression of enhanced green fluorescent protein (EGFP) primarily to adult mouse testis. Specifically, robust EGFP expression (both transcript and fluorescence) is observed in adult mouse testis in a developmentally-regulated, stage-specific expression pattern during spermatogenesis. Much lower EGFP transcript and fluorescence is observed in adult ovaries. Very low levels of EGFP transcript expression (but not fluorescence) is reported in fetal and newborn gonads of both sexes. These Dazl-eGFP transgenic mice may be useful for gametogenesis studies, including pachytene spermatocytes.
The Dazl-eGFP transgene was designed with a 1.7 kb mouse Dazl (deleted in azoospermia-like) sequence (located upstream of the translational start site that also included the 5' UTR) all upstream of an enhanced green fluorescent protein (EGFP) coding sequence and SV40 polyA sequence. This transgene was microinjected into the maternal pronucleus of fertilized mouse FVB/N oocytes. Founder mice (line 10-54) were subsequently mated with wildtype FVB/N mice to establish the colony. Dazl-eGFP mice were bred with wildtype FVB/N mice for approximately 7 generations prior to arrival at The Jackson Laboratory. Upon arrival, transgenic mice were bred with FVB/NJ (Stock No. 001800) to establish the colony.

When maintaining a live colony, hemizygous mice may be bred to wildtype siblings or to FVB/NJ inbred mice. The donating investigator reports that homozygous mice are viable and fertile.
