A loxP-flanked puromycin resistance/STOP cassette followed by a fusion of firefly luciferase and synthetic antigen peptide SIYRYYGL were introduced to the Gt(ROSA)26Sor (gene trap ROSA 26, Philippe Soriano) locus. After Cre activity, the recombined locus has high expression of Luciferase-SIY fusion protein. This mutant mouse strain may be useful in studies of immune response to a self-antigen or over-expressed tumor-associated antigen when used in combination with tumor-prone models.

Targeted (Reporter) | Gt(ROSA)26Sor | gene trap ROSA 26, Philippe Soriano |
Mice that are homozygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. In the absence of Cre, there is low level expression of Luciferase-SIY H2b antigen expression. After Cre activity, the recombined locus has high expression of Luciferase-SIY fusion protein. This mutant mouse strain may be useful in studies of immune response to a self-antigen or over-expressed tumor-associated antigen when used in combination with tumor-prone models. This mouse is also useful as a reporter for Cre activity.
A loxP-flanked puromycin resistance/STOP cassette followed by a fusion of firefly luciferase and synthetic antigen peptide SIYRYYGL were introduced to the targeted gene using 129S4/SvJae-derived J1 embryonic stem (ES) cells. This line was made and maintained on a 129S genetic background.
Expressed Gene | luc, luciferase, firefly |
---|---|
Site of Expression | When these mutant mice are bred to mice that express Cre recombinase, the resulting offspring will overexpress the luciferase-SIY fusion protein in the cre-expressing tissues. |
Allele Name | targeted mutation 3, Tyler Jacks |

Allele Type | Targeted (Reporter) |
Allele Synonym(s) | Gt(ROSA)26Sortm2Tyj; R26LSL-LSIY |
Gene Symbol and Name | Gt(ROSA)26Sor, gene trap ROSA 26, Philippe Soriano |
Gene Synonym(s) | |
Promoter | CAG, CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter, |
Expressed Gene | luc, luciferase, firefly |
Site of Expression | When these mutant mice are bred to mice that express Cre recombinase, the resulting offspring will overexpress the luciferase-SIY fusion protein in the cre-expressing tissues. |
Strain of Origin | 129S4/SvJae |
Chromosome | 6 |
Molecular Note | A cassette containing a CAG promoter, a floxed STOP followed by a luciferase gene and sequence encoding the antigen SIYRYYGL (SIY) was inserted into the locus. Cre-mediated recombination is necessary to allow for expression of luciferase/SIYRYYGL under the control of the CAG promoter. |

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
