These transgenic mice express an isoform of the human insulin-like growth factor 1 (somatomedin C), (IGF-1des), under the control of the minimal -426/+28 rat probasin (Pbsn) promoter. Hemizygous transgenic male mice develop spontaneous atypical prostate hyperplasia, but the lesions do not progress to neoplasia within 1 year. This mutant mouse strain may be useful in studies of prostate tumorigenesis.

These transgenic mice express an isoform of the human insulin-like growth factor 1 (somatomedin C) under the control of the minimal -426/+28 rat probasin promoter. The expressed des(1-3) IGF-1 isoform (IGF-1des) has decreased interaction with IGF binding proteins due to a 3-amino acid deletion in the N terminus of the mature protein. The transgene is detected in the dorsolateral and ventral prostate lobes by RT-PCR analysis. Hemizygous transgenic male mice develop spontaneous atypical prostate hyperplasia, but the lesions do not progress to neoplasia within 1 year. Histological analysis reveals lesions consistent with mouse prostatic intraepithelial neoplasia (PIN) with increased abnormal acini. Female mice that are homozygous for the transgene are viable, normal in size and do not display any gross physical or behavioral abnormalities. This mutant mouse strain may be useful in studies of prostate tumorigenesis.
When bred to C57BL/6-Tg(TRAMP)8247Ng/J mice (see Stock No. 003135 ), the double transgenic mice initially exhibit slower temporal progression
of pathology, but then eventually develop more
advanced adenocarcinomas and more widespread metastatic deposits than single transgenic TRAMP mice.
A transgenic construct containing the human insulin-like growth factor 1 (somatomedin C) isoform, IGF-1des (that has a 3-amino acid deletion in the N terminus of the mature protein), under the control of the minimal -426/+28 rat probasin promoter, and carrying SV40 polyadenylation site sequences was injected into fertilized FVB/N mouse eggs. Founder line 5305 was established. The mice were backcrossed to FVB/N mice for 10 generations. The Donating Investigator maintained the mice as hemizygotes.

When maintaining a live colony, these mice can be bred as homozygous females to hemizygous males. The Donating Investigator did not routinely breed to homozygosity and maintained the strain as a hemizygote.
