These mice carry a threonine to alanine (T140A) phosphorylation site mutation in exon 3 of the activation-induced cytidine deaminase gene. Homozygotes are developmentally normal. B cells are impaired but not ablated in their ability to undergo class switch recombination and somatic hypermutation.
Exon 3's threonine 140 was mutated to alanine (bp change is A95G) and a loxP-flanked neomycin resistance cassette was place in intron 2. A B6(Cg)-Tyrc-2J/J-derived embryonic stem cell line was used to create the mutation. The neomycin cassette was excised with Cre, leaving a single loxP site. A C57BL/6 genetic background was maintained by the donating laboratory.
