A tandem series of adenine/thymine (A/T) repeats were introduced to an Igk (immunoglobulin Κ Vk167) transgene. This "TgA" line has an A/T sequence comparable to that found in line "TgG" without the single cytosine:guanine (C:G) insert (see Stock No. 008719) and does not show any increase in number of mutations in the activated B cell A/T repeat region of 4-12 month old mice. This strain may be useful in studying somatic hypermutations associated with the rapid diversification of antibodies during immune responses to antigen.

Alternating EcoRV and PvuII sites and a tandem series of adenine/thymine (A/T) repeats with a single guanine:cytosine (G:C) base pair were introduced to an Igk (immunoglobulin kappa; VΚ167) transgene. The cytosine (C) residue marks a site at which deamination to uracil (U) can occur, initiating somatic hypermutations of upstream and downstrean A/T residues. This "TgA" line has an A/T sequence comparable to that found in line "TgG" without the single cytosine:guanine (C:G) insert (see Stock No. 008719
) and does not show any increase in number of mutations in the activated B cell A/T repeat region of 4-12 month old mice. This strain may be useful in studying somatic hypermutations associated with the rapid diversification of antibodies during immune responses to antigen. Hemizygotes are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities.
VΚ167/PEPS, indirectly derived from BALB/c myeloma protein MPOC 167, was designed to carry alternating EcoRV and PvuII sites (EPS) in a transgene with a truncated region upstream of the promoter.
The Igk (immunoglobulin kappa) sequence was further modified in the current strain through the insertion of 5 tandem repeats of a 20-bp adenine/thymine (A/T) fragment. This "TgA" line has an A/T sequence comparable to that found in line "TgG" without the single cytosine:guanine (C:G) insert (see Stock No. 008719).
The transgene was initially introduced to a C57BL/6-SJL background, and subsequently crossed to BALB/cByJ or C57BL/6 inbred mice for a total of 8 generations by the donating investigators.
