HRC transgenic mice express a transgene encoding a T cell receptor (TCR) αβ heterodimer specific for histone H4 (86-92) peptide VVYALKR presented by the H-2Kb class I MHC molecule. The TCRαβ transgene is expressed on >90% of mature T cells. Mice that are hemizygous for the transgene are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. This mouse strain may be useful for studies of immune regulation, cancer, and autoimmunity.

Rearranged V α2-SGTGGYKV Tcra and V α8.3-SGTGGSAETL Tcrb chains recognizing histone H4 (86-92) peptide VVYALKR were identified in a population of CD8+ T cells infiltrating mouse prostate tumors. These were cloned into separate targeting constructs. Linearized sequences were co-injected into (CBAxC57BL/6)F2 embryos and resultant mice were backcrossed more than ten times to C57BL/6 by the donating laboratory. This line gives "complete" expression of the transgenic TCR αβ heterodimer on >90% of CD8α+ cells.
