These Gabrb3 (gamma-aminobutyric acid (GABA-A) receptor, subunit beta 3) conditional (floxed) mutant mice may be useful in generating conditional mutations for studying the role of neurodevelopmental diseases and sensitivity to anesthetics and ethanol.

These mice possess loxP sites on either side of exon 3 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissue(s).
When bred to a strain expressing Cre recombinase in the neuronal cells (see Stock No. 003966 for example), this mutant mouse strain may be useful in studies of neurodevelopmental diseases.
When bred to a strain expressing Cre recombinase in the hippocampal CA1 pyramidal cell layer (see Stock No. 005359 for example), this mutant mouse strain may be useful in studies of neurodevelopmental diseases.
Of note, several strains bearing gamma-aminobutyric acid (GABA-A) receptor mutations are available from this donating investigator (Dr. Gregg Homanics, University of Pittsburgh), including Gabra1 (Stock No. 004318), Gabra4 (Stock No. 006874), Gabra6 (Stock No. 002710), Gabrb3 (Stock No. 002711 and Stock No. 008310), Gabrd (Stock No. 003725), and Gabrg2 (Stock No. 003137).
A FRT site flanked targeting vector containing the neomycin resistance gene was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 3 of the targeted gene, and another loxP site was inserted upstream of exon 3. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected in C57BL/6J blastocysts. Resulting chimeric male animals were backcrossed to wildtype C57BL/6J female mice. The mice were then crossed to the FLP recombinase expressing strain B6;SJL-Tg(ACTFLPe)9205Dym/J (Stock No. 003800) to excise the neo cassette. The Tg(ACTFLPe)9205Dym transgene was bred out of the strain by backcrossing the mice to C57BL/6J for one additional generation. The mice retain one FRT site and loxP sites flanking exon 3.
