Description

These mice possess loxP sites flanking exons 3 through 6 of the targeted gene. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 3 through 6 deleted in the cre-expressing tissue(s). This mutant mouse strain may be useful in studies of neurological development.

Detailed description

These mice possess loxP sites flanking exons 3 through 6 of the targeted gene. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Histological analysis of brains from homozygotes reveals slightly wider CA1 and CA3 regions and a split in CA2 region in the hippocampus. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 3 through 6 deleted in the cre-expressing tissue(s).

Development

A loxP site flanked targeting vector containing PGK-neo was utilized in the construction of this mutant. This selection cassette was inserted into intron 2 of the targeted gene, and another loxP site was inserted into intro 6. This construct was electroporated into 129S6/SvEvTac derived TC-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to 129S6/SvEvTac mice for 9 generations. Upon arrival at The Jackson Laboratory, mice were bred to 129S1/SvImJ mice (Stock No. 002448) for at least one generation to establish the colony.

Allele

Symbol: Pafah1b1^tm2Awb
Name: targeted mutation 2, Anthony Wynshaw-Boris
MGI accession ID: MGI:2384060
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed); Hypomorph
Synonyms: Lis1-loxP; Pafah1b1^loxPex3-6; Pafah1b1-loxP
Marker symbol: Pafah1b1
Marker name: platelet-activating factor acetylhydrolase, isoform 1b, subunit 1
Marker synonyms: Lis1; LIS-1; LIS2; lissencephaly-1 protein; MDCR; MDS; Mdsh; NudF; PAFAH; PAF-AH 45; Pafaha
Chromosome: 11
Strain of origin: 129S6/SvEvTac
Molecular note: A floxed PGK-neo cassette was inserted into intron 2 and a third loxP site was introduced into intron 6 via homologous recombination. This allele is hypomorphic, because of disruption of transcription or splicing by the insertion of a phosphoglycerate kinase neomycin cassette from the targeting vector into the second intron (J:82268).
