These Gbx2flox mice harbor loxP-flanked homeodomain portion of exon 2 of the gastrulation brain homeobox 2 (Gbx2) locus and may be useful in generating conditional mutations for studying gastrulation brain homeobox protein function in the embryonic mesencephalon and rhombomere 1, as well as developing cerebellum and thallamus.

Mice homozygous for the Gbx2flox conditional allele are viable and fertile, with loxP sites flanking the protein coding region of exon 2 of the targeted gene. When bred to mice that express Cre recombinase, the resulting offspring will have this region (encoding the gastrulation brain homeobox 2 homeodomain) deleted in the cre-expressing tissue(s). These Gbx2flox mice may be useful in generating conditional mutations for studying gastrulation brain homeobox protein function in the embryonic mesencephalon and rhombomere 1, as well as developing cerebellum and thallamus.
A targeting construct was designed to insert an frt-flanked neomycin cassette and loxP site in intron 1, and a loxP site downstream of the protein coding region within the second exon of the targeted gene. This construct was electroporated into 129S6/SvEvTac-derived W4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts and chimeric males were bred with C57BL/6 females. The resulting Gbx2flox-neo mice were next bred to mice constitutively expressing Flpe recombinase (129Sv genetic background) to remove the frt-flanked neo cassette. The resulting Gbx2flox offspring were selectively bred to Swiss Webster mice to remove the Flpe transgene, and then maintained by breeding to Swiss Webster mice and also interbred to generate homozygous mice prior to arrival at The Jackson Laboratory. Upon arrival, Gbx2flox mice were bred to 129S1/SvImJ (Stock No. 002448) for at least one generation to establish the colony.
