Mice homozygous for this transgene are viable and fertile and do not display any gross physical or behavioral abnormalities. The mice express humanized Renilla Green Fluorescent Protein (hrGFP, Stratagene) specifically in germ cells of the testes under the control of the Dazl (deleted in azoospermia-like) promoter. Expression is highest in pachytene spermatocytes. No expression was found in the liver, heart, small intestine, large intestine, spleen, or kidney of these mice. This strain may be useful in studies of male fertility.
A 1.7 kb promoter fragment from just upstream of the translational start site of Dazl was amplified from B6SJLF1 genomic DNA by PCR. The PCR product was ligated into PCR Blunt (Invitrogen). The promoter fragment was cut out of PCR Blunt using NsiI and NotI, and then ligated into the NsiI and NotI sites of phrGFP (Stratagene) to generate pDazl-hrGFP. The Dazl-hrGFP construct was cut out of pDazl-hrGFP free of most other vector sequence using NsiI and MluI and used for pronuclear injection of B6SJLF1 oocytes to generate transgenic lines. This is founder line 4.
