EGFP under the control of the Sry promoter is expressed in fetal gonads similarly to endogenous Sry, first in cells located centrally in the genital ridge and then later in cells located at the cranial and caudal poles. EGFP is expressed exclusively in pre-Sertoli cells in the urogenital ridge.
Tg(Sry-EGFP)92Ei was generated via random insertion after micro-injection into the pronucleus of C57BL/6J fertilized eggs. The resulting transgenic strain, C57BL/6J-Tg(Sry-EGFP)92Ei/EiJ (Stock No. 003927), was bred to homozygosity and a female was bred to a male C57BL/6J-Chr YAKR/J/EiJ (Stock No. 007250) when that consomic was at generation N56. The progeny were backcrossed to homozygous C57BL/6J-Tg(Sry-EGFP)92Ei females until generation N15. In 2007 sperm were cryopreserved from a homozygous male at generation N15F2.

The construct contains two SV40 early mRNA polyadenylation signal in place of the endogenous Sry 3' UTR. Expression of EGFP indicates that the spatial expression of EGFP mRNA and protein is similar to endogenous Sry. |

Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
