Description

F5^PltTg transgenic mice (also called Pf4A or Pf4fvA) express mouse F5 (coagulation factor V) cDNA from the mouse Pf4 (platelet factor 4; also called Cxcl4) promoter. RT-PCR indicates expression limited to cells of megakaryocyte/platelet lineage in the spleen and bone marrow. Though a component of the transgenic construct, EGFP expression is not detectable.

Detailed description

F5^PltTg transgenic mice (also called Pf4A or Pf4fvA) express mouse F5 (coagulation factor V) cDNA from the mouse Pf4 (platelet factor 4; also called Cxcl4) promoter. RT-PCR indicates expression limited to cells of megakaryocyte/platelet lineage in the spleen and bone marrow. Though a component of the transgenic construct, EGFP expression is not detectable.

When combined with an F5 knock-out allele (Stock No. 004078) to produce homozygous knock-out/hemizygous transgenic animals, mice show a slightly increased bleeding time, a platelet F5 level that is ~5% that of wildtype mice, no plasma F5, and transgene rescue of the perinatal hemorrhage phenotype associated with F5 knock-outs.

Development

The mouse Pf4 (platelet factor 4; also called Cxcl4) promoter drives expression of full-length mouse F5 (coagulation factor V) cDNA in this transgenic line. An internal ribosome entry site sequence (IRES) followed by an enhanced green fluorescent protein (EGFP) coding sequence and a bacterial kanamycin resistant gene (KAN) were also incorporated downstream, but expression of EGFP is not detected. A chimeric intron with optimized splice signals from the pCI expression vector was introduced upstream of the F5 cDNA in the transgene cassette to enhance gene expression. The vector was injected into fertilized oocytes created by mating B6SJLF1 or C57BL/6 female mice with B6SJLF1 male mice. This line has been backcrossed minimally (perhaps twice) to C57BL/6 by the donating laboratory.
