Description

This strain contains loxP sites flanking exon 2 of the wingless-related MMTV integration site 4, Wnt4, gene. These Wnt4-floxed mice may be useful to generate Cre-conditional mutations for studying sex determination, female reproductive biology (oocyte development and ovarian follicle biology), adipogenesis, limb and joint development, growth plate development, skeletogenesis, kidney development, and tumorigenesis. In addition, mutations in this gene are associated with Mayer-Rokitansky-Kuster-Hauser syndrome and SERKAL syndrome in humans.

Detailed description

This strain contains loxP sites flanking exon 2 of Wnt4 resulting in a Cre-dependent conditional null allele. Homozygotes are normal. Studies by Kobayashi et al., determined that when this conditional allele is exposed to Cre expression by Amhr2^tm3(cre)Bhr Mullerian duct regression proceeds normally.

Development

Exon 2 was flanked by loxP sites and and an FRT flanked neomycin resistance cassette was inserted downstream of exon 2 in 129S7/SvEvBrd-Hprt⁺ derived AB1 ES cells, from which mice were generated. The resulting line was crossed to a strain bearing Gt(ROSA)26Sor^tm1(FLP1)Dym to excise the neomycin resistance cassette, resulting in the Wnt4^tm1.1Bhr allele, which still has exon 2 intact. This was imported into The Jackson Laboratory in 2006 for Dr. Eva Eicher and in 2007 sperm was cryopreserved from heterozygous males at generation N2F2 to C57BL/6J.
