These floxed mutant mice possess loxP sites flanking the exon that encodes amino acids 236-258 of the Man2a1 gene. This strain may be useful for generating conditional mutations in applications related to human congenital dyserythropoietic anemia type II.

Mice homozygous for this floxed targeted mutation are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. When bred to Cre transgenic strains, the loxP-flanked exon is recombined and deleted where Cre is expressed, producing a null allele.
A targeting vector was used to place a floxed neomycin-thymidine kinase expression cassette downstream of the exon that encodes amino acids 236-258 and an additional loxP site upstream of the exon. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were electroporated with Cre expression plasmid pCre-Hygro to excise the neo fragment and leave the floxed exon. Gancyclovir-resistant ES cells were injected into C57BL/6 blastocysts. The line as provided has been backcrossed only once to C57BL/6 by the donating laboratory.
