These targeted reporter mutant mice express PER2::LUC bioluminescence similar to endogenous Per2 expression, and may be useful as a real-time reporter of circadian dynamics in different tissues.

Mice homozygous for this "mPer2Luc" mutation are viable and fertile with no developmental or morphological differences compared to wildtype littermates. Expression of PERIOD2::LUCIFERASE (or mPER2::LUC) fusion protein during peak periods is similar to endogenous pPER2 patterns. Homozygous mice have normal entrainment and circadian behaviors. These mPer2Luc knock-in mice may be useful as a real-time reporter of circadian dynamics in different tissues.
A targeting vector was designed to insert the firefly luciferase (luc) gene followed by an in-frame loxP-flanked neomycin resistance gene between exon 23 and the 3' UTR of the period circadian clock 2 (Per2) gene. The construct was electroporated into 129S6/SvEvTac-derived W4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. Chimeric males were bred to C57BL/6J females. Heterozygotes were backcrossed to C57BL/6J inbred mice for 7-11 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
