Description

This IRES-tau-mCherry knockin reporter bicistronically co-expresses tau-mCherry fusion protein with the endogenously expressed transient receptor potential cation channel (Trpc2) and is valuable for expression analysis in the olfactory and vomeronasal systems.

Detailed description

Transient receptor potential cation channel (Trpc2) is essential in chemosensory signal transduction in mouse vomeronasal sensory neurons, and is also expressed in two types of sensory neurons of the mouse main olfactory epithelium (Omura and Mombaerts, 2014). This knockin allele has an internal ribosome entry site and the microtubule-binding protein tau/mCherry (taumCherry) fusion protein downstream of the Trpc2 coding sequence to produce bicistronic expression. Vomeronasal and olfactory sensory neurons that express Trpc2 also co-express the axonal marker taumCherry fusion protein by virtue of IRES-mediated co-translation, permitting single-cell characterization of Trpc2 expressing live neurons.

Development

The Trpc2^tm2Mom mutation was generated in 129P2/OlaHsd derived E14 ES cells and resulting mice were bred to C57BL/6J before being intercrossed to homozygosity. This mutation was generated using the ACNf neo cassette, which self-excises during transmission in the male chimera. Sperm was cryopreserved from homozygous males on this mixed B6;129P2 background.
