This targeted reporter has bicistronic expression of the endogenous Olfr749 and tauGFP. GFP expression is found sparsely distributed in the dorsal main olfactory epithelium at 9 days of age and embryonic immunohistochemistry shows a steady increase in the number of olfactory sensory neurons (OSNs) expressing GFP from E15.5 to birth. There are approximately 95 GFP positive OSNs at embryonic day 15.5, approximately 500 at birth, and approximately 8,450 at 24 days of age. At embryonic day 15.5 the axons of these OLFR749 OSNs extend to the protoglomerular layer in the medial olfactory bulb, by E16.5 they form a web-like appearance, the formation of discrete medial glomeruli is seen after birth, and the medial glomerular diameter increases from perinatal day 1 through 24. This allele has been used to characterize the onset of prenatal odorant responsiveness by olfactory sensory neurons bearing OLFR749. Patch-clamp recordings showed that by embryonic day 16.5 these olfactory sensory neurons respond to 2-phenylethyl alcohol.
This targeted insertion was generated in 129P2-derived E14 ES cells and after an outcross to C57BL/6J this strain was maintained by sibling breeding on this mixed background. Sperm was cryopreserved from homozygous males.
