Description

Olfactory sensory neurons that express the olfactory receptor Or13c7 (formerly Olfr155) also co-express the tauGFP fusion protein by virtue of IRES-mediated co-translation. These GFP reporter mice are useful in studies to identify olfactory sensory neuronal development and topography in glomeruli in the olfactory bulb.

Detailed description

Olfactory sensory neurons that express the olfactory receptor Or13c7 (formerly Olfr155) also co-express the tauGFP fusion protein by virtue of IRES-mediated co-translation. These neurons can be revealed by the intrinsic fluorescence of GFP, or by immunofluorescence with anti-GFP antibodies. This reporter allele is combined with other olfactory receptor reporter alleles in Stock Number 027730.

Development

This targeted mutation was generated in 129P2/OlaHsd derived E14 ES cells and the resulting mice from clone H26 were crossed to Tg(EIIa-cre) bearing mice to excise the loxP flanked pgkneo (LNL) cassette. Although this transgene was generated in FVB/N mice, it was backcrossed onto C57BL/6J prior to this cross. The offspring were intercrossed and the transgene was selectively bred out of this line. Thus, this STOCK background is predominantly C57BL/6J and 129P2/OlaHsd. In 2007 sperm were cryopreserved from homozygous males.

Molecular note: The gene was tagged by insertion of an IRES-GFP-floxed neo cassette 3' to the stop codon. Transient expression of cre recombinase in correctly targeted ES cells removed the floxed neo cassette prior to blastocyst injection. Immunofluorescence was observed in mature olfactory sensory neurons of mutant animals.
