Description

These transgenic SCID mice express the human histocompatibility complex (MHC) class I HLA-A2.1 allele.

Detailed description

Although transgenic NOD mice develop diabetes significantly earlier than do NOD/ShiLtDvs (NOD) mice, this transgenic strain harbors the Prkdc^scid mutation (characterized by an absence of functional T cells and B cells) and therefore does not become diabetic. In adoptive transfer experiments splenocytes from NOD.HLA-A2.1 mice did not induce T1D in these NOD-scid.HLA-A2.1 transgenic mice at a significantly faster rate than when transferred to NOD-scid recipients. This strain is useful for adoptive transfer experiments and as a source of insulitis free pancreatic islets.

Development

NOD.CB17-Prkdc^scid was crossed once with NOD.B6-TgN(HLA-A2.1)1Enge/Dvs (Stock #003088) and the doubly heterozygous offspring were backcrossed once to NOD.CB17-Prkdc^scid to make the strain homozygous for Prkdc^scid then sibling mated to fix the strain homozygous for TgN(HLA-A2.1)1Enge as well.

Molecular note: The transgene contains the full length HLA-A2.1 gene. Expression of human class I MHC Ag HLA-A2.1 was detected on cells from homozygous transgenic spleen, bone marrow, and thymus. Line 1 inserted into the gene at 18736683-18757058 (Build GRCm38/mm10) resulting in a duplication. The duplication results in a functional knock-out. Founder line 1 has a copy number of 3-8.
General note: Expression of this human class I molecule in transgenic mice does not result in expansion of the number of cytotoxic T lymphocyte (CTL) precursors specific for other human class I Ag, HLA-B27 or HLA-A2.2.
