Description

These mutant mice contain a "Lox-STOP-Lox" cassette that prevents expression of the human NOTCH1 transgene until excised by Cre-recombinase. This strain may be useful for generating conditional mutations in applications related to the study of notch signaling during apoptotic cell death.

Detailed description

Transgenic mice are viable, fertile and behaviorally normal. These "CALSL-NICD (H)" mice (or simply CALSL-NICD) reportedly carry 10-20 copies of the transgene inserted into a single genomic locus. Expression of the transgene-derived intracellular domain of human NOTCH1 is prevented by a "Lox-STOP-Lox" cassette. When transgenic mice are bred to a strain expressing Cre recombinase, the "floxed stop" cassette is excised in the resulting offspring, and human NOTCH1 expression is observed in the cre-expressing tissue(s). These transgenic mice may be useful in studying early neural progenitor cell development and apoptosis, and responses to tissue-specific Notch activation.

For example, when crossed to a strain expressing Cre recombinase in the nervous system (see Stock No. 003771), this transgenic mouse strain may be useful in studies of notch signaling during apoptotic cell death.

Development

The human Notch1 intracellular domain (NICD) (amino acid residues 1762–2304; including the RAM23 domain, CDC10/Ankyrin repeats, and two nuclear localization signals), as well as a SV40 late polyadenylation signal was inserted downstream of a chicken beta-actin-loxP-stop-loxP sequence. The transgene was injected into the pronucleus of C57BL/6J embryos. The resulting transgenic mice with 10-20 copies of the transgene at a single insertion site (called CALSL-NICD (H) or simply CALSL-NICD), were subsequently bred to C57BL/6J mice for many generations prior to arrival at The Jackson Laboratory.

Molecular note: This transgene consists of the chicken beta-actin promoter followed by a loxP flanked stop, the human NOTCH1 intracellular domain (amino acids 1762-2304), and an SV40 late polyadenylation signal. Cre mediated recombination removes the stop and allows expression of the NOTCH1 gene. Only single site insertions were used to generate mice. This line carries 10 - 20 copies of the transgene and protein is detectable by western blot analysis after cre mediated recombination.
